After entering the Ticket for the WoW64 thing I awaited the 1.0.79.0 installer to work which it seems it does. (Full Install including GAC) Thanks for that.
But on Win7 X64 with VS2010 I am still unable to create a new DataSource (New DatabaseConnection) since there is nothing with SQLite as DataSource as it was in Win7 X86 or using the old 1.0.66.0
I have no clue what is missing to have SQLite back to the DataSource possibilities.

You'll need to install the 32-bit setup package because Visual Studio is 32-bit only.
